    The present study was undertaken in order to examine transduction mechanism involved in the single application of 100 μM homocysteine (Hcy) on isolated rat femoral artery (RFA) rings equilibrated on the basal tone; to establish if a single application of 100 μM Hcy alters contractile effect of phenylephrine (Phe), or oppositely the relaxant effect of acetylcholine (ACh) or bradykinin (BK) after 60-min-long incubation of 100 μM Hcy; and finally to identify morphological changes on the vascular wall after a 24-h-long incubation of 100 μM Hcy. Hcy produced contractile response of intact RFA, which was increased after endothelial denudation, while decreased by urapidil (an α1 receptor blocker), nifedipine (a voltage-gated L-type Ca++ channel blocker) or indomethacin (a cyclooxygenase inhibitor). The initial RFA contraction evoked by Phe was further increased by the single addition of Hcy, which was not the case when ouabain (an inhibitor of Na+/K+-ATPase) was preincubated. After 60-min-long incubation of Hcy relaxant actions of ACh and BK were unaltered, equieffective and equipotent. A 24-h-long incubation of RFA rings with Hcy produced an impairment of vascular endothelium, expressed as a minor or more pronounced interruption of endothelial cells. © 2014 Akadémiai Kiadó, Budapest.

    The aim of our study was to investigate the effects of ifenprodil and MK-801 on D,L-homocysteine thiolactone induced seizures in adult rats.Male Wistar rats were divided into following groups: 1. Saline-treated (C, n=10); 2. D,L-homocysteine thiolactone 8 mmol/kg, i.p. (H, n=7); 3. Ifenprodil 20 mg/kg i.p. (IF, n=8); 4. MK-801 0.5 mg/kg, i.p. (MK, n=8) and 5. Groups that received IF or MK 30 minutes prior to H (IFH, n=8 and MKH, n=8). Seizure behavior was assessed by incidence, latency, number and intensity of seizure episodes. Seizure severity was described by a descriptive scale with grades 0-4. Lethality in experimental group was recorded 90 min and 24 h upon D,L-homocysteine thiolactone administration.There were no behavioral signs of seizure activity in groups C, IF and MK.Pre-treatment with MK-801 (MKH) showed tendency to reduced incidence of convulsions, latency to the first seizure onset and the severity of seizure episodes, but statistical significance was not attained comparing to the H group. However, median number of seizure episodes was significantly decreased in MKH (p<0.05), comparing to the H group. On the other hand, ifenprodil (IFH) decreased the latency to the first seizure onset and increased the median number of seizure episodes (p<0.05). The majority of seizure episodes in IFH (72.1%, p<0.05) and MKH (73.1%, p<0.05) groups was grade 2 and significantly different comparing to the H (36.0%). Our findings suggest that MK-801 has a mild anticonvulsive effect on D,L-homocysteine thiolactone induced seizures in adult rats. © 2011 Akadémiai Kiadó, Budapest.

    Some side-effects of excessive physical training are ascribed to reactive oxygen species production. In this work we investigated the effects of progressively imposed maximal physical effort (levels I to V), using progressive maximal exercise test, on peripheral blood lactate, NO (through NO 2-), superoxide anion (O2-) and methemoglobin (MetHb) in a group of 19 elite soccer players. Blood lactate (mmol/L) was increased (4.55, level V vs. resting level, 1.95). The basal production of NO2- was in the direct relation with O 2 consumption. Significant increase (p<0.05) in O2 - values at effort level I (4.18) as compared to the resting value (4.01), and the significant increase (p<0.01 or p<0.05) in the MetHb (%) was found between II (18.79) and III (19.63) or between II and IV (19.24) effort levels, respectively. The regression lines of NO2- and O2- crossed at the level of the respiratory compensation point (RC), suggesting that RC could be of a crucial importance not only in the anaerobic and aerobic metabolism but in mechanisms of signal transductions as well. The results could be of the theoretical interest and also useful in designing an athlete training strategy. © 2011 Akadémiai Kiadó, Budapest.
